BACKGROUND背景

Notherapiesarecurrentlyapprovedforthetreatmentofpulmonaryhypertensioninpatientswithinterstitiallungdisease.ThesafetyandefficacyofinhaledTreprostinilforpatientswiththisconditionareunclear.

目前还没有批准用于治疗间质性肺病患者的肺动脉高压的疗法。吸入曲前列素治疗这种疾病的安全性和有效性尚不清楚。

METHODS方法空白

Weenrolledpatientswithinterstitiallungdiseaseandpulmonaryhypertension(documentedbyrightheartcatheterization)inamulticenter,randomized,doubleblind,placebo-controlled,16-weektrial.Patientswereassignedina1:1ratiotoreceiveinhaledtreprostinil,administeredbymeansofanultrasonic,pulsed-deliverynebulizerinupto12breaths(total,72μg)fourtimesdaily,orplacebo.Theprimaryefficacyendpointwasthedifferencebetweenthetwogroupsinthechangeinpeak6-minutewalkdistancefrombaselinetoweek16.SecondaryendpointsincludedthechangeinN-terminalpro–B-typenatriureticpeptide(NT-proBNP)levelatweek16andthetimetoclinicalworsening.

RESULTS结果

Atotalofpatientsunderwentrandomization,withassignedtoinhaledtreprostinilandtoplacebo.Baselinecharacteristicsweresimilarinthetwogroups.Atweek16,theleast-squaresmeandifferencebetweentheTreprostinilgroupandtheplacebogroupinthechangefrombaselineinthe6-minutewalkdistancewas31.12m(95%confidenceinterval[CI],16.85to45.39;P0.).Therewasareductionof15%inNT-proBNPlevelsfrombaselinewithinhaledtreprostinilas
